It doesn't get "watered down" in successive generations, even if both parents don't have it; it either of a person's parents is a Detian, that person will be as well. The only functional difference for a "first generation" recipient is that they weren't born that way.
The one exception I can think of is Tali'Shukra vas Venachar's family line, where it appears to have skipped a generation, but then she received a treatment that was reverse-engineered for quarians from a stolen sample of human Omega-2 that had already been revised for salarians, so who the fuck knows. :)
